When the environment temperature is not suitable for the growth of fruit flies (temperature is lower than 18 ℃), within 7 days after the medium is inoculated, it is particularly easy to grow mold.

To avoid mold growth, you should pay attention to:

① Vessels for cultivating fruit flies should be strictly sterilized. After washing the culture bottle, let it dry naturally, and then dry sterilize with a cotton plug at high temperature (120 ℃, 30min).

â‘¡ The nutrition of the culture medium should be comprehensive. There are many types of Drosophila culture medium. The corn medium used in this laboratory is as follows: 750mL of water is boiled, 6.2g of agar is added, and 62.5g of white sugar is added after the agar is dissolved. Corn paste (corn flour 82.5g, adjusted to a paste with cold water), boil for 2 to 3 minutes, add 7g of yeast powder, heat for 1 to 2 minutes, stir well, remove the fire source, add 2mL of propionic acid The role of mildew). After the medium has cooled slightly, pour it into a sterilized medium bottle, try not to let the medium stick to the wall of the bottle, plug the cotton plug, and put it in a drying oven with residual heat to evaporate the water on the wall of the bottle .

③ Eliminate poison during inoculation. Inoculate in a sterile room. If there is no sterile room, wipe your hands with a 75% alcohol cotton ball before inoculation. After inoculation, put them in an incubator suitable for the growth temperature of fruit flies (temperature 20 ~ 25 ℃). Once the next generation of adult flies are hatched, the medium is not easy to grow mold.

Treatment method after mold growth:

â‘ If the culture medium grows mold, the mold spores float in the entire space of the culture flask, including the fruit fly. If the fruit fly is inoculated into a new medium, the medium will grow out again after about 4 to 5 days. When the mold comes, there are only two solutions to the problem: one is to stop inoculating this moldy fruit fly and re-inoculate the non- moldy fruit fly; the second is to take measures to kill the mold spores on the fruit fly.

â‘¡The fungicidal method is: pour the adult fly with mold spores into an empty culture bottle, pour an appropriate amount of 75% alcohol on the cotton plug, and then tightly close the bottle mouth, using alcohol is volatile, and the volatile alcohol fills the entire The bottle will kill the mold spores on the fruit fly. Two points should be noted in operation: first, the amount of alcohol poured on the tampon should be appropriate, too many fruit flies are prone to drunk and faint, and too little can not kill mold spores; second, the time the fruit flies stay in the empty bottle , 2d is better.
